where exactly on the back should i drill it? Measurements?
It really depends on what you want to do with the holes and the placement of it. Like if you want a sump under your stand you should probably go with two holes on the bottom of the back of the tank and on different ends of the back. Hope that makes sense but im not that good with wording things. What size bulkheads are you going to use?

always drill your holes near the top as there is less pressure near the top of the glass, i would say 4" from the side and 5" down would be perfect
and the size of holes depends on what size of return pump your planning on putting on it
i would go with 1 1"ID bulk head with a mag7 or 9 as your return pump the more holes you put in your tank the more junk you will have cluttering your tank

+1^; Not only that, but if you put them at the bottom and your return pump shuts off for some reason, your going to have a major flood as all the water drops to your sump, and lots of dead organisms as they have no water. I personally would drill the drain hole a 1 1/2", and place a elbow so it points up at an angle to within a short distance of the top (you only want it to drain the water level to the bottom of your plastic strip, otherwise it always looks like you are need to add water to your tank). The return size can be smaller (depending on the size of hose or pvc you run from the return pump). be sure and put a backflow preventer on your return line, or again, major flood. Also cover the drain elbow with a sponge or something to prevent snails from crawling into it and plugging it up. Good luck.

easy way to regulate the leavel of water in your tank is to make a overflow box, easy to make look at a after marker drilled tank some time and youll see what i mean,
make a 3 sided box and cut little slits in the top of it about 1/4 and make sure that the slits dont go more then 1 1/4 down or like Scopus Tang said it will look like you need to add water all the time
a way i keep my sump from over flowing when i shut the pump off or the power gose out is drill a hole right under the water surface of the return tube so when the water drains past that hole it will suck air and break the syphon,
