You will need to start the spiphon over, sounds like you put the air tube too far in to the U tube when you started to spiphon, insert it only to the bend, and make sure you get all the air out of it. Good Luck

Yeagh, just suck out the air bubble as much as you can. I find that bubbles that are the size of a quarter get flushed out once the water gets flowing. The trick is to not let air get back in when you pull the airline from the U tube. I usually just keep my mouth on it until I have it removed.

Once you prime the overflow's U Tube with the airline, suckingn out the the air, you should never have to reprime the U tube if it's not removed from the system.
If you select a pump that has a higher flowrate, the water will force all air bubble out of the U tube.
You need to have a return pump that delivers enough water to the tank, that will drive out these accumulating air bubbles, while at the same time not exceed the flow capacity of your overflow.
A balanced system will never have air bubbles, will always start upon loss of power, and not flow more than the system will allow.
If you shut off your pump and remove the U Tube, naturally you must reprime the tube.
What pump are you using ?
What is the rating on your overflow ?
What size is your tank ?
