I'm going to set up a ??gallon rubber-made tub for a auto top-off. My questions is, does the water need to be aerated while in the tub or does it matter? Thanks,

In my best estimation - yes
The top off water in the holding tank should be heated and aerated with a powerhead .. ESPECIALLY if it's RO or RO/DI water that you are making at home.
Allowing dissolved CO2 gas in the freshwater to reach equalibrium with the CO2 gas in the atmosphere ( room ) is an important step.
Heating and aerating the top off water would be a great thing to do ;)

Necessary - possibly
Absolutely necessary - nope
It all depends on your set up and individual conditions ;)
