how to drill a hole acrylic??

To drill a hole in acrylic without any chance of damage, you can use a dremel moto tool. Use the grinder bit. If I don't have a hole saw the size I need, I just draw the size of hole I want, drill a small hole in the middle and then use the dremel to enlarge the hole.
