I can tell you from experance I have done it befor but it was thick acrylic if you can do it at 1 i cant see why 2 would be a problem?

Guiding the holesaw can be tricky as there is nothing to hold the pilot bit in place.
What I like to do when I do this is to take a piece of scrap acrylic (or wood if in a pinch) that is slightly larger than the size of the new hole. Drill a hole through it with the holesaw you intend to use. Then take a small file and slightly enlarge the hole so the holesaw moves freely (rather than snugly) through the hole.
Then tape that down over the old hole using strong tape. That will form a guide to help your holesaw stay in one place.

or you can go to harbor freight, and get a 2" step drill bit, just go slowly and you'll have no problems
