when I am building the dividers should I just use a good strong plexiglass? Will that be sufficient enough?

Yes. I got acrylic plates from Home Depot (~$15 total for all dividers and cover plates).
I would make the design and assure yourself of the dimensions of each divider. Ask Home Depot to cut the plates (dividers) for you. They did mine free of charge with the purchase of the plates. You can do your own cutting, but you have to be careful.
This worked great with a square glass tank. If you get a plastic container, you have to see if the sides of the container are straight up or not. If it isn't straight, it will be harder to cut the dividers.
