Water has nothing to do with cycling. Matter of fact water from a cycled tank would be worse than freshly mixed saltwater because it would be of worse water quality than new water and may have free floating parasites or other nasty stuff. It's the bacteria on the live rock and sand that keeps...

Considering that 99% of store bought stands have nothing but the 3/4 inch sides for strenth and those even come with warrenties you would be more than fine with 2X4's.

You need to test for iodine and then dose if you want to. You should never dose anything without testing first even if the label gives a dpsing amount.
